No supports are needed for printing. If your printer isn't well-calibrated, consider slightly scaling up the retainers. They are designed to fit tightly to prevent the ruler from skewing.
For two-color printing with a single extruder, change the filament for the top layers to get a different color.
To print a 220x220 ruler on a 220x220 bed, simply disable the skirt by setting skirt loops to 0.
